Answer:

Your uncle is full of it. Aging is not caused by one single disease and there are no universal diseases.
Most humans carry E. coli in their intestines but it's a symbiont in it's natural habitat and actually helps digestion; it's only dangerous when it moves higher up in the digestive system and acquires certain mutations.
Most humans also have reoviruses in their system, but these viruses cause no symptoms and are not related to aging.
Aging is caused by a lot of things. Mitochondria wear out, telomeres get shortened, neurons and other cells with no associated somatic stem cells die and are never replaced…
The problem is that at age 40 you're off your warranty. There was a time when 35 was about as old as anybody got due to lack of medical treatment, sabretooth tigers, neighboring tribes, famines, that sort of thing. There are places where the average lifespan is still in the high 30s. No one lived long enough to die of Alzheimer's or congestive heart failure.
Humans are still evolving but it's mostly things like the disappearance of genes for excessive sweat glands. We keep inventing wheelchairs and heart bypass surgeries and fertility treatments which slow down evolution, and some day we’ll have genetic engineering to do it's job for it.
